The use of leaf and internode explants and different combinations of growth regulators for callus formation, regeneration and root formation of black bean plant (Hyoscyamus niger L.)

Abstract
In recent years, callus formation, regeneration and in vitro fertilization of medicinal plants with genetic engineering purposes are one of the interesting topics in biotechnology and plant tissue culture. In line with this goal, the optimization of the regeneration of the medicinal plant, Hyoscyamus niger L., was carried out in vitro in a factorial manner in the form of a completely randomized design with 3 treatments in 3 replications. The test factors include the type of micro sample (leaf and internode), different growth regulators that include 3 treatments: one (T1) containing 1 mg/L BA and 0.5 mg/L 2,4-D, treatment two ( T2) included NAA and BA each at the rate of 5 mg/l and treatment three (T3) contained 3 mg Kin and 1 mg 2,4-D. T0 (hormone-free) treatment was used as control. The results showed that 100% callus formation occurred in the leaf explant with treatment 2 and in the stem internode microsample in treatment 3, the highest fresh weight of callus and the largest diameter of callus produced in leaf tissue occurred in treatment T1. The highest number of regeneration and stem formation, 100%, was observed in the internode tissue of the stem in T2 treatment. In the T3 treatment, 87% of the calli produced in the leaf entered the regeneration stage and 92% of the stem internode samples were regenerated. In T2 treatment, new stems had better longitudinal growth. Root production was observed in stem internode explants and in leaves and directly in T3 treatment.
