تحلیل توپولوژیکی شبکه‌های متابولیتی، الگوی مقاومت گندم مقاوم به ویروس موزائیک رگه‌ای گندم را در شرایط تنش دمایی آشکار می‌کند

چکیده
ویروس موزائیک رگه‌ای گندم (wheat streak mosaic virus=WSMV) از خانواده پوتی‌ویریده (Potyviridae) از مهمترین ویروس‌های آلوده کننده گندم در جهان و ایران محسوب می‌شود. در این تحقیق تجزیه و تحلیل شبکه متابولیتی مربوط به آنالیز LC-qTOF-MS در رقم مقاوم کراس عدل، ۲۴ ساعت پس از مایه‌زنی در دماهای بهینه (C°20) و بالا (C°32) با استفاده از نرم افزار تحت وب Metaboanalyst 6.0 مورد بررسی قرار گرفت. با سنجش پارامترهای شبکه مشخص شد که پاسخ‌های متابولیکی سیستمیک نسبتاً متفاوتی در رقم مقاوم آلوده به WSMV در دماهای بهینه و بالا وجود دارد. در دمای بهینه، افزایش بیان نوکلئوتیدها از قبیل آدنین، گوانین و اسفینگولیپیدها (اسفینگنین) مشاهده شد که در دفاع ضد ویروسی از طریق القای مرگ برنامه‌ریزی شده در تجمع ROS نقش دارند. از طرفی کاهش اسیدهای آمینه آروماتیک (فنیل آلانین و تریپتوفان) دیده شد که احتمالاً منابع کربن را به سمت مسیرهای دفاعی هدایت می‌کنند. تحت تنش دمای بالا کاهش میزان ترکیباتی از قبیل کولین (تخریب غشای سلولی در دمای بالا) و ۲-آمینوبنزوئیک اسید (به عنوان مهارکننده ویروس) و افزایش اسید آمینه‌های پرولین (افزایش تحمل گیاه به دمای بالا) و آرژینین (افزایش دفاع مبتنی بر خاموشی آر.ان.ای و سنتز پروتئین های مرتبط با تنش میزبان) رخ داد. این پژوهش نشان داد که مقاومت به WSMVنتیجه هماهنگی دقیق شبکه‌های متابولیتی چند بعدی در مبارزه با چندین تنش همزمان مثل بیمارگر و دمای بالا می‌باشد. از یافته های بدست آمده می‌توان برای توسعه ارقام با مقاومت پایدار در برابر تنش‌های زیستی و غیرزیستی در شرایط تغییر اقلیم استفاده کرد.
