بررسی واکنش آنتی‌اکسیدانی سیانوباکتر Nostoc sp.به­عنوان یک کود زیستی در تنش اسمزی ناشی از پلی‌اتیلن گلیکول پس از پرتودهی گاما

چکیده
مقدمه: سیانوباکتری Nostoc sp. به‌دلیل توانایی تثبیت نیتروژن، تولید متابولیت‌های زیست‌فعال و بهبود حاصلخیزی خاک، به‌عنوان یک کود زیستی ارزشمند در کشاورزی پایدار شناخته می‌شود. بااین‌حال، تنش اسمزی خاک یکی از مهم‌ترین تنش‌های غیرزیستی است که رشد و عملکرد این کودهای زیستی را به شدت محدود می‌کند. یکی از رویکردهای نوین برای افزایش تحمل به تنش، استفاده از پیش‌تیمارهای فیزیکی نظیر پرتوتابی گاما به منظور القای مکانیسم‌های دفاعی و ایجاد تحمل متقاطع است. پژوهش حاضر با هدف بررسی این فرضیه طراحی شد که پرتوتابی گاما می‌تواند از طریق فعال‌سازی پیشگیرانه سیستم آنتی‌اکسیدانی، مقاومت Nostoc sp. را به تنش اسمزی افزایش دهد. بدین منظور، تغییرات سطح گونه‌های فعال اکسیژن (ROS) و فعالیت آنزیم‌های سوپراکسید دیسموتاز (SOD) و پراکسیداز (POD) در پاسخ به اثر متقابل پرتوتابی گاما و تنش اسمزی مورد ارزیابی قرار گرفت.
مواد و روش‌ها: آزمایش به‌صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار اجرا شد. فاکتور اول شامل پرتوتابی گاما در شش سطح دُز (۰، ۲۰، ۵۰، ۱۰۰، ۵۰۰ و ۱۰۰۰ گری) به‌عنوان پیش‌تیمار و فاکتور دوم شامل تنش اسمزی بود. سطح ROS با روش فلوریمتری و پروب DCFH-DA اندازه‌گیری شد. فعالیت SOD بر اساس مهار احیای NBT و فعالیت POD با روش اسپکتروفتومتری و سوبسترای پیروگالول سنجیده شد. داده‌ها با آنالیز واریانس دوطرفه و آزمون توکی در سطح معنی‌داری 01/0 تحلیل گردیدند.
یافته‌ها: نتایج نشان داد که اثر اصلی پرتوتابی، اثر اصلی تنش اسمزی و اثر متقابل آن‌ها بر هر سه شاخص بیوشیمیایی در سطح 01/0 معنی‌دار بودند. در نمونه‌های بدون پیش‌تیمار پرتوتابی، اعمال تنش اسمزی منجر به افزایش سطح ROS ( تا حدود ۱۷ برابر شاهد) و متعاقب آن کاهش فعالیت SOD و POD در دُزهای بالای تنش اسمزی گردید. اما پرتوتابی پیش‌گیرانه با دُزهای متوسط (۵۰ و ۱۰۰ گری) به طور معنی‌داری از افزایش ROS جلوگیری کرد. قابل توجه‌ترین یافته آن بود که در نمونه‌های پیش‌تیمار شده با دُز ۱۰0۰ گری، فعالیت SOD در شرایط وجود تنش از 13/31 به 41/92 واحد افزایش یافت (افزایش حدود 3 برابری) و فعالیت POD نیز از 45/12 به 03/74 واحد رسید (افزایش حدود 6 برابری). دُزهای بالای پرتوتابی (۵۰۰ و ۱۰۰۰ گری) به تنهایی اثرات مخربی داشتند، اما در ترکیب با تنش اسمزی، نمونه‌های پیش‌تیمار شده با دُز ۱۰۰ گری بهترین عملکرد حفاظتی را نشان دادند.
بحث و نتیجه‌گیری: یافته‌های این پژوهش نشان می‌دهد که پرتوتابی گاما با دُز (۵۰ و۱۰۰گری) می‌تواند به‌عنوان یک راهکار مؤثر برای افزایش مقاومت به تنش اسمزی در سیانوباکتری Nostoc sp. به کار رود. این پدیده از طریق مکانیسم تحمل متقاطع توجیه می‌شود. پرتوتابی خفیف با القای سطح پایه ROS و فعال‌سازی پیشگیرانه آنزیم‌های SOD و POD، سلول را برای مقابله با افزایش شدید ROS ناشی از تنش اسمزی آماده می‌کند. به عبارت دیگر، پرتوتابی «سیستم هشدار اولیه» را فعال کرده و آستانه تحمل سلول را جابه‌جا می‌سازد. این یافته پیامدهای عملی مهمی برای تولید کودهای زیستی مقاوم به خشکی دارد. پیش‌تیمار گاما می‌تواند به‌عنوان یک فرآیند صنعتی ساده و کم‌هزینه، کارایی Nostoc sp. را در خاک‌های شور که بخش قابل توجهی از اراضی کشاورزی را تشکیل می‌دهند، بهبود بخشد. این پژوهش پایه علمی برای توسعه نسل جدیدی از کودهای زیستی پرتوتابی شده با قابلیت تحمل به تنش‌های چندگانه محیطی فراهم می‌آورد.
